<summary type="text">Characterization of Polyphenol Oxidase from Phyllanthus Amarus (Pokok Dukung Anak) Leaves

Recently much attention has been paid to biologically active components of herbs, in this study the polyphenol oxidase (PPO) was extracted, purified and characterized from herb Phyllanthus amarus, the most popular medicinal herb in South East Asia. It was extracted and purified by ammonium sulphate (NH4)2SO4 precipitation, isolated by ion-exchange chromatography and purified by gel filtration chromatography and HPLC. The biochemical analysis revealed that the PPO extracted from Phyllanthus amarus leaves has higher affinity towards catechol (KM = 17.4mM and Vmax = 22,077 U/ml min-1) at an optimum pH of 7.1. The enzyme had an optimum temperature of 390C and was relatively stable up to 560C for a period of 90 minutes with almost 80% activity remaining. Among the various PPO inhibitors tested, ascorbic acid was found that it is the most effective inhibitor for the enzyme with 10mM catechol as substrate.

Stroke in pregnancy may cause substantial risks and effects to the mother and foetus. Pregnancy related ischaemic stroke following arterial occlusion had been reported to be more common compared to pregnancy associated haemorrhagic stroke.&#13;
Various underlying conditions and factors may contribute towards the risk of developing ischaemic stroke in pregnancy. Inevitably, ischaemic stroke in pregnancy would have a significant impact on the antenatal and obstetric care of the patients.We present a case report to illustrate a rare case of ischaemic stroke occurring in the second trimester of pregnancy. A 36 years old lady, gravida 3 para 2 with no omorbidities was diagnosed with left middle cerebral artery infarction at 16 weeks period of amenorrhea which was relatively early compared to the usual presentation of ischaemic stroke at third trimester or postpartum. Investigations to ascertain the possible underlying causes and associated risk factors did not reveal any &#13;
significant findings. She was prescribed with aspirin until 38 weeks period of gestation with no bleeding complication reported.

This review article highlights the management and treatment options of hyperthyroidism especially in Malaysia and discusses the contemporary utilisation of radioiodine therapy emphasising on the fixed dose regimen.

A healthy, 26-year-old Chinese gentleman presented with sudden onset of reduced vision in the right eye associated with floaters. He claimed to be lethargic for the past three weeks prior to presentation. There was no history of other constitutional&#13;
and systemic symptoms. He has no significant family history of malignancy or eye problem. Examination revealed right best corrected visual acuity (BCVA) of 6/36 and left BCVA was 6/9. Anterior segment examinations were normal. However, funduscopy revealed bilateral hyperemic optic discs swelling, dilated and tortuous retinal veins with multiple dot and blot haemorhages including Roth spots. There was also vitreous haemorhage in the right eye. The other cranial nerves were normal. Systemic examination revealed no organomegaly. Blood investigations were suggestive of Chronic Myloid Leukemia (CML) in chronic phase.
